P2.06.57 TIM-1+ B Cells Promote Immune Suppression and Predict Poor Prognosis in Lung Adenocarcinoma

This study investigates the role of TIM-1 positive B cells (TIM-1B cells) in lung adenocarcinoma (LUAD) and their impact on patient prognosis and immune suppression. Using multiplex immunofluorescence on tissue samples from 126 LUAD patients, the researchers assessed TIM-1 distribution, mature tertiary lymphoid structures (TLS), and infiltration of CD8+ T cells in both primary tumors and tumor-draining lymph nodes (TDLN).<br /><br />Key findings include that high levels of TIM-1B cells in tumors and TDLN correlate with poorer disease-free survival and overall survival in LUAD patients. TIM-1B cells were shown to exert immunosuppressive functions by hindering the maturation of TLS and reducing the infiltration of cytotoxic CD8+ T cells within tumors. Negative correlations were observed between TIM-1B cell proportions and TLS maturation (r = -0.3990) and between TIM-1B cells and CD8+ T cell infiltration (r = -0.3928).<br /><br />These results suggest that TIM-1B cells promote an immunosuppressive tumor microenvironment that facilitates tumor progression. Consequently, TIM-1B cells may serve both as a valuable prognostic biomarker to predict patient outcomes in LUAD and as a potential therapeutic target to enhance anti-tumor immunity.<br /><br />The authors emphasize that targeting TIM-1B cells could improve immune responses by promoting TLS maturation and increasing CD8+ T cell infiltration, ultimately enhancing treatment efficacy. This study adds to existing knowledge on TIM-1 signaling’s role in regulating immune cells, particularly B cells, within the tumor microenvironment, highlighting its promise for future research and clinical applications in lung adenocarcinoma.<br /><br />In summary, TIM-1B cells contribute to immune suppression and poor prognosis in LUAD by disrupting TLS development and CD8+ T cell-mediated anti-tumor immunity, making them a critical focus in therapeutic strategies.
