EP02.01. Androgen Receptor Suppresses Lung Cancer Invasion and Cisplatin Resistance via Decreasing TPD52 Expression - PDF(Abstract)

This abstract discusses a study on the role of androgen receptor (AR) in lung cancer invasion and cisplatin resistance. The researchers aimed to explore the mechanisms by which AR impacts the progression of non-small-cell lung cancer (NSCLC), particularly its connection to TPD52-mediated cell invasion and resistance to cisplatin.<br /><br />The study used various methods such as plasmid construction, MTT assay, Transwell invasion assay, Western blot assay, qRT-PCR assay, chromatin immunoprecipitation assay (ChIP), luciferase reporter assay, in vivo studies, and statistical analysis.<br /><br />The results showed that AR can suppress NSCLC cell invasion and cisplatin resistance by decreasing the expression of a circular RNA called circ-SLCO1B7. AR directly binds to the 5' promoter region of its host gene SLCO1B7 to suppress circ-SLCO1B7. This circ-SLCO1B7 acts as a sponge to inhibit the function of miR-139-5p, a micro RNA. The inhibition of miR-139-5p leads to increased translation of Tumor protein D52 (TPD52) messenger RNA by reducing miRNA binding to its 3' untranslated region (3'UTR).<br /><br />The researchers confirmed these findings using an in vivo mouse model with orthotopic xenografts of NSCLC cells, as well as analysis of human NSCLC databases and clinical specimens.<br /><br />Overall, the study suggests that AR plays a role in suppressing NSCLC cell invasion and drug resistance by modulating the AR/ circ-SLCO1B7/ miR-139-5p/ TPD52 signaling pathway. Targeting this signaling pathway may lead to the development of new treatment strategies to better suppress NSCLC progression.
