EP02.02. Identification of COL5A2+ Cancer-Associated Fibroblast in Solid Subtype of LUAD by Integrated Analysis of WGCNA and Single Cell RNA-seq - PDF(Abstract)

This research study aimed to investigate the factors contributing to poor prognosis in the solid subtype of lung adenocarcinoma (LUAD). The researchers used integrated analysis of weighted gene co-expression network analysis (WGCNA) and single-cell RNA sequencing (RNA-seq) to identify a specific type of cancer-associated fibroblast (CAF) associated with poor prognosis in this LUAD subtype. The researchers also explored the communication and differentiation of cells in the tumor microenvironment using the CellChat toolkit and trajectory analysis. <br /><br />Using the WGCNA approach, the researchers identified a key module highly associated with the solid pattern of LUAD, which represented extracellular matrix organization. They then determined a group of hub genes, including COL5A2, THBS2, COL4A2, COL6A2, and COL15A1, using protein-protein interaction network analysis. Further analyses using multiple datasets confirmed that COL5A2 was significantly increased in the solid predominant subtype of LUAD and strongly associated with poor prognosis in patients with lung adenocarcinoma.<br /><br />The researchers analyzed a single-cell RNA-seq dataset and found that COL5A2 was exclusively expressed in a cluster of CAF derived from the solid predominant subtype of LUAD. These COL5A2-expressing CAFs also expressed chemokines, including CXCL10 and CCL19. In the analysis of cell communication, the COL5A2 CAFs were found to interact with multiple immune cells, such as proliferating T cells, B cells, macrophages, and plasmacytoid dendritic cells. Patients with high levels of COL5A2 CAFs showed resistance to immunotherapy.<br /><br />Overall, this research suggests that COL5A2 CAFs play a crucial role in the poor prognosis of the solid pattern LUAD. The findings highlight the importance of understanding the tumor microenvironment and cell interactions for developing effective therapeutic strategies for this subtype of lung adenocarcinoma.
