EP03.05. RNA Sequencing to Characterize Pathways in Patients with EGFR-mutated Non-Small Cell Lung Cancer - PDF(Slides)

Researchers at Rush University Medical Center conducted a study to analyze gene expression pathways in patients with EGFR-mutated non-small cell lung cancer (NSCLC) and determine any differences based on age at diagnosis. Using RNA sequencing, the study evaluated 281 patients identified through the Tempus Lens database.<br /><br />The researchers performed a differential gene expression analysis using comprehensive RNAseq data, focusing on patients below and above 70 years old. Enhanced volcano plots were generated to visualize gene expression differences using the edgeR package in Bioconductor. Additionally, a Gene Set Enrichment Analysis was conducted using the KEGG reference database to identify modulated genes.<br /><br />The results showed significant differences in gene expression and upregulated pathways between the two age groups. Patients above 70 years old at diagnosis exhibited an upregulation of pathways related to cancer cachexia and inflammation. Interestingly, the study also found a general upregulation of cortisol and aldosterone synthesis and secretion, along with processes associated with Cushing's syndrome in patients above 70 years old. Furthermore, downregulation of lipid mobilization and carbohydrate metabolism (glycolysis and gluconeogenesis) was observed.<br /><br />The study concluded that RNA sequencing is a promising tool to translate genomic features into clinical aspects of EGFR-mutated lung cancer. By stratifying patients based on age, researchers were able to identify significant differences in gene expression and pathways associated with age-related factors. These findings contribute to a better understanding of the molecular mechanisms involved in cancer development and may have implications for personalized treatment approaches in EGFR-mutated NSCLC.
