EP03.06. Differential Anticancer Compound Response of Filipino Patient-derived Lung Adenocarcinoma Cells (GL01) Against A549 Cell Line - PDF(Abstract)

This study aimed to derive primary lung adenocarcinoma (LUAD) cells from a Filipino patient and compare them to the commonly used A549 cell line in terms of cell viability and migration. The researchers successfully immortalized the LUAD primary cells (GL01) and observed that they formed monolayers with epithelial morphology, similar to A549 cells. The MTT assay results showed that GL01 cells had significantly lower cell viabilities when treated with guyabano extracts (GE) at certain concentrations compared to 5-fluorouracil, a commercial antineoplastic drug. However, A549 cells showed significantly lower cell viabilities when treated with 5-fluorouracil, indicating that the commercial drug is less effective in the GL01 cells. In the wound healing assay, GE inhibited cell migration in GL01 cells at a specific concentration, but no inhibition was observed in A549 cells. These findings suggest that LUAD cells from different origins may exhibit variable phenotypes and responses to treatments. The authors suggest that the genetic background could play a role in the differential anticancer drug response and may have implications in cancer drug discovery and personalized medicine. Overall, this study highlights the importance of using diverse cell lines, especially those derived from patients of different ethnic backgrounds, to better understand tumor biology and develop more tailored therapeutic strategies.
