EP04.01. METTL7A Inhibits Cell Proliferation and M2-Like Polarisation of Tumor-Associated Macrophage in Lung Adenocarcinoma - PDF(Slides)

A study conducted by researchers from Fujian Medical University and Fujian Cancer Hospital in China has identified a protein called METTL7A that acts as a tumor suppressor in lung adenocarcinoma (LUAD). The researchers found that low levels of METTL7A are associated with malignant progression and poor prognosis in LUAD patients. Using gene expression analysis and survival prognosis models, they demonstrated that METTL7A expression is correlated with longer overall survival and progression-free survival.<br /><br />Further analysis revealed that METTL7A plays a role in regulating immune response and T cell activation pathways, while inhibiting spliceosome and cell cycle pathways. In experimental studies, overexpression of METTL7A in LUAD cells inhibited cell proliferation, migration, and M2-polarization of tumor-associated macrophages (TAMs). In vivo studies using a xenograft model confirmed the tumor-suppressive effects of METTL7A overexpression.<br /><br />The researchers also examined the distribution of METTL7A in the tumor microenvironment and found that it was mainly present in mono/macro cells, epithelial cells, and malignant cells. METTL7A expression was positively correlated with various immune cell types. The findings suggest that METTL7A may modulate the tumor microenvironment and contribute to the generation of M2 TAMs.<br /><br />Based on these findings, the researchers propose that METTL7A could serve as a potential biomarker for LUAD diagnosis and prognosis. They suggest that targeting METTL7A and its effects on TAM polarization could be a viable strategy for LUAD treatment.<br /><br />The study was supported by grants from the National Natural Science Foundation of China and the Natural Science Foundation of Fujian Province. The researchers acknowledge the TCGA and GEO working groups for providing data for their analysis.
