P1.07. Patient-Derived Xenografts from Thoracic Malignancies as Promising Preclinical Models - PDF(Abstract)

Researchers from Kumamoto University Hospital have developed patient-derived xenograft (PDX) models from thoracic malignancies, which could serve as promising preclinical models for studying cancer. Precision oncology, a new clinical approach for cancer patients, requires appropriate preclinical tools to develop personalized treatment strategies. PDX models, generated by engrafting patient tumors into immunodeficient mice, are seen as valuable tools for preclinical and clinical research. The Kumamoto University Lung Cancer Patient Derived Xenograft (KLC-PDX) project was initiated to create PDX models for thoracic malignancies with fewer treatment options. Tumor tissue acquired from surgical resection and biopsy was xenografted into immunodeficient mice, and similarities between the PDXs and patient tumors were analyzed. The researchers found good concordance in histology and molecular profiles between patient and PDX generations, and drug efficacy tests showed different sensitivities among the same histological type, similar to actual clinical patients. The PDXs were successfully passaged to more than the 5th generation, and major gene mutation information was maintained up to the 3rd generation. RNA-seq analysis revealed certain trends of biological changes in chemo-naïve and chemo-resistant PDXs, indicating inter-patient and intra-tumor heterogeneity. The researchers concluded that the developed KLC-PDX model could recapitulate tumor heterogeneity and be applied in various stages of cancer diagnostics and therapeutics. Overall, this study highlights the potential of PDX models in advancing precision oncology by providing a better understanding of tumor heterogeneity and personalized treatment approaches.
