Researchers from Lucence Diagnostics have developed an ultrasensitive next-generation sequencing assay using AmpliMark technology to detect lung cancer. The study focused on plasma cell-free RNA (cfRNA), an analyte that has been relatively unexplored compared to cell-free DNA (cfDNA) in liquid biopsy assays. The researchers designed an cfRNA expression panel targeting 24 mRNA and long non-coding RNA markers, with cfRNA abundance estimated using the geometric mean read counts of housekeeping genes and the RNA expression score obtained by summing the relative normalized expression of all RNA targets. <br /><br />The study analyzed plasma cfNA samples from 40 patients, including 24 with lung cancer (37.5% early-stage) and 16 non-cancer controls. The researchers found that cfRNA levels were elevated in early stages of lung cancer compared to non-cancer controls. They also observed that positive cancer-associated cfDNA and cfRNA markers increased with cancer stage, and the expression of mirrored expression markers, which are presumptively tumor-associated and absent in non-cancer controls, increased with lung cancer stage. <br /><br />The researchers concluded that mirrored expression markers profiling provides an independent and potentially earlier measure of cancer signals compared to cfDNA profiling. They also suggested that mirrored expression markers could be a reliable analyte to complement cfDNA-based technologies for sensitive and early detection of lung cancer. However, further evaluation of the study is warranted to confirm the findings. <br /><br />Overall, this study demonstrates the potential of cfRNA sequencing for sensitive detection of lung cancer and highlights the importance of exploring cfRNA as an analyte in liquid biopsy assays.
