P1.24. A Comparative Analysis of Secreted MiRNAs Reveals Candidate Biomarkers for Pleural Mesothelioma Detection - PDF(Abstract)

Researchers have conducted a comparative analysis of secreted miRNAs to identify potential biomarkers for the detection of pleural mesothelioma (PM). MicroRNAs released by tumor cells have been found to play a role in tumor growth and treatment resistance. However, little is known about the miRNAs secreted in PM. <br /><br />The researchers established primary cell cultures from pleural effusion of PM and non-PM patients. They profiled two types of secreted miRNAs: total secreted miRNA in cell culture supernatant and exosomal miRNAs. They identified differentially expressed miRNAs using DESeq2 and conducted survival analysis using the survival package. Additionally, they selected miRNA target genes using MirDB and functionally annotated them using DAVID.<br /><br />The analysis revealed 309 deregulated miRNAs in PM exosomes and 84 in the supernatant, with miR-16-5p, let-7c-3p, miR-23a-3p, and miR-30a-5p among the up-regulated miRNAs. MiR-30a-5p was found to be a significant predictor of patient survival. The researchers also identified potential target genes of the up-regulated miRNAs, which were associated with transcriptional regulation and cell division.<br /><br />This study highlights the importance of the PM secretome in understanding mesothelioma biology and discovering novel biomarkers. The researchers identified 11 miRNAs as potential biomarkers for PM. The selective packaging of miRNAs into exosomes suggests their potential use as cell-free biomarkers. Further analysis is currently underway to validate these findings.<br /><br />Overall, this research provides valuable insights into the role of secreted miRNAs in PM and the potential for using them as biomarkers for early detection.
