P2.22. Single-nuclei RNA Sequencing Reveals Heterogeneity and Malignancy in Pulmonary Sarcomatoid Carcinoma - PDF(Abstract)

Researchers from Shanghai Pulmonary Hospital and Tongji University School of Medicine conducted a study aimed at understanding the specific cell atlas and single-cell heterogeneity of pulmonary sarcomatoid carcinoma (PSC), a rare and aggressive form of non-small cell lung cancer. They used single-nuclei RNA sequencing (snRNA-seq) to decipher the PSC-specific regulatory signatures. The study involved 11 frozen, untreated surgical tumor tissues including 5 PSC, 3 lung adenocarcinomas (LUAD), and 3 lung squamous cell carcinomas (LUSC). <br /><br />Through snRNA-seq analysis, the researchers identified seven major populations among all samples, including epithelial cells, macrophages, fibroblast-like macrophages, lymphocytes, fibroblasts, and endothelial cells. They also found 23,000 cancer cells with a high copy number variation (CNV) score. The researchers defined 11 cancer cell states, with some states primarily originating from PSC and others mainly existing in LUAD and LUSC. Notably, they discovered a cell type with high plasticity and identified several enriched signaling pathways in this cell type.<br /><br />Using non-negative matrix factorization (NMF), the researchers extracted seven intratumor meta-programs with co-expressed genes. Two of these programs, enriched in the 'Proliferation,' 'Matrix,' and 'Plasticity' states, demonstrated a significant prognostic value in LUAD and LUSC based on data from The Cancer Genome Atlas (TCGA) database.<br /><br />Overall, the snRNA-seq analysis provided a high-resolution transcriptional map and in-depth knowledge of the tumor heterogeneity of PSC. This study contributes to a better understanding of PSC and may have implications for the development of targeted therapies for this aggressive form of lung cancer.
